
Further characterization of the photoreaction center from Ectothiorhodospira sp. detection of the H subunit by monoclonal antibodies

The photoreaction center isolated from Ectothiorhodospira sp. is known to contain polypeptides L,M and C (a four heme c cytochrome) in a 1:1:1 molar ratio. As a test for the presence of a H subunit, Western blots of the chromatophore and photoreaction center polypeptides were reacted with a monoclonal antibody directed against the H subunit of Rhodospirillum rubrum . The chromatophores but not the isolated photoreaction center cross‐reacted with this antibody. It is concluded that the H polypeptide is lost during the purification of the photoreaction center which, in its native state, therefore contains four polypeptides including a genuine H subunit.
