The pel/ptsM locus codes for two apparently unrelated activities: transport of mannose by the phosphoenolpyruvate‐dependent phosphotransferase system (PTS) and penetration of bacteriophage λ‐DNA. This locus is known to consist of at least two genes, ptsL ( manX ) and pel/ptsM ( manYZ ). In this paper we present the cloning and analysis of the manXYZ locus. Subcloning and complementation analysis of the different fragments suggest that the locus contains 3 separate genes manX, manY and manZ and that the corresponding gene products are involved in mannose uptake. In addition, manY codes for the Pel function, necessary for penetration of λ‐DNA.
