
Neutralisation of biological activities of pertussis toxin with a monoclonal antibody

Three monoclonal antibodies (L5, L7 and L10) against pertussis toxin (PT) were produced by the fusion of myeloma cells with spleen cells of mice immunised with pertussis toxoid and PT. Immunoblotting showed that each monoclonal antibody bound to more than one subunit of the toxin. L5 and L7 bound predominantly to subunit S3 and with much less affinity to subunit S1, while L10 bound mainly to subunit S1. L7 strongly neutralised the lymphocytosis‐promoting and histamine‐sensitising activities of PT, whereas L5 and L10 showed no or only partial neutralising effect on these activities. The strongest neutralisation of PT activity in haemagglutination and Chinese hamster ovary (CHO) cell assays was also found with L7.
