A new procedure for introducing plasmid DNA into Streptomyces lividans protoplasts by electroporation is described. Transformation of S. lividans by plasmid DNA was observed under various conditions by altering the electric pulse voltage, number of pulses per cycle, cycle time, and number of cycles. The maximum number of transformants was obtained under conditions in which some lysis of the protoplasts was observed. The maximum transformation frequency obtained using electroporation was 10 4 /μg of plasmid pVE28.
