Lipopolysaccharide (LPS) was recovered from the aqueous layer of phenol‐extracted defatted Paracoccus denitrificans cell envelopes, although the presence of 3‐deoxyoctulosonic acid in interfacial and phenol‐soluble materials indicated that the LPS was relatively lipophilic. LPS contained phosphorus, 3‐deoxyoctulosonic acid, glucose, galactose and glucosamine. Heptose was absent. The 4 fatty acids present were identified, by gas‐liquid chromatography‐mass spectrometry (GLC‐MS) as ester‐linked 3‐OH‐10:0 and 12:1 and amide‐linked 3‐OH‐14:0 and 3‐oxo‐14:0. Chemical analyses, sodium dodecyl sulphate‐polyacrylamide gel electrophoresis (SDS‐PAGE) and fractionation studies showed the LPS to be an R‐type. The occurrence of an R‐type LPS is compatible with the permeability properties of the outer membrane. The hydroxy and oxo fatty acids and their linkages, the lack of heptose, and the R‐type LPS, further emphasize the close phylogenetic relationship between P. denitrificans and Rhodobacter capsulata and Rhodobacter sphaeroides .
