Cloning and DNA homology of 3‐isopropylmalate dehydrogenase genes from thermophilic bacilli

The genes coding for 3‐isopropylmalate dehydrogenase from the moderate thermophile Bacillus caldotenax and the extreme thermophile Bacillus caldolyticus were cloned in Escherichia coli using pBR322 as a vector. The restriction maps of the cloned fragments were determined and the small fragments carrying the enzyme gene were subcloned. DNA‐DNA hybridization of DNA fragments containing the gene from mesophiles ( Bacillus subtilis, E. coli and Saccharomyces cerevisiae ) and thermophiles (Bacillus coagulans, B. caldotenax, B. caldolyticus and Thermus thermophilus ) were examined. The DNA fragments from the thermophiles hybridized with each other, but not with the mesophile DNA fragments.
