Lipopolysaccharides which are masked by the presence of a large capsule in many Gram‐negative organisms such as Escherichia coli K29, can be localized in ultrathin sections of fixed and Lowicryl‐embedded cells. The ‘post‐embedding‐immuno‐labeling’ procedure revealed a layered organization of the antigens, with the O‐9 specific lipopolysaccharide at the outer membrane's surface and the extensive K29 polysaccharide coat encapsulating the organism.
