Restriction enzyme analysis of mitochondrial DNA (mtDNA) appears to be a useful technique for the examination of the possible relationship of organisms [1]. Using this method several studies have already been carried out on such groups as Aspergillus [2], Paramecium [3] and rats [4]. In this paper we have used restriction enzyme analysis to compare the mtDNAs of a number of strains of the free-living protozoa belonging to the genus Acanthamoeba. Such a study would be of some value as previous attempts to examine the relationships of these organisms using morphological characters have not been wholly successful.
