A NEW METHOD AND INSTRUMENT FOR THE QUANTITATIVE DETERMINATION OF CHLOROPHYLL

Thespectrophotometric methoddepends onmeasuring the-width ofthe absorption bandsofchlorophyll intheredendofthespectrum. This methodisneither speedy norpractical formakingaconsiderable number ofdeterminations, since foreachdetermination a separate spectrophoto- graphmuLst betaken, whichrequires considerable time.Inaddition, an expensive spectrograph isnecessary, andtheaidofanexpert physicist is advisable forcorrectly interpreting thephotographs. Likethecolorimetric method, itispossible toascertain concentrations ofchlorophyll onlytotwo decimal places. Thepurpose indeveloping theinstrument heredescribed wastoelimi- natethepossibilities ofpersonal error inobservation, todetect moreminute differences thanispossible bythepreceding methods, andtomakethe quantitative determination ofchlorophyll aneasier andsimpler process. Thisinstrument willdetect differences of5mg.between samples, andhas amaximumerror of9percent. atthelowerconcentrations andaminimunm errorof2percent. atthehigher concenitrations, whicherrors inmostcases affect onlly thethird decimal place. Figures 1and2illustrate theapparatus.
