Arabinogalactan proteins are abundant O ‐glycosylated proteins in plants and important in many cellular processes. The glycosylation processes have been believed to occur in the secretory pathway through endoplasmic reticulum ( ER ) and Golgi, however, we identified arabinogalactan glycosyltransferases target to yet uncharacterized subcellular compartments as well as Golgi cisternae. Colocalization and pharmacological studies implicate the uncharacterized compartments likely to be a part of the so‐called unconventional protein secretion pathway, thus indicating the presence of multiple routes for glycoprotein secretion in plants.
