structure and modifies its functions in metabolic homeostasis related to energy metabolism and catabolism of isoleucine branched-chain fatty acids. Thus, the blockade of ABAD/Ab interaction using ABAD-DP may be a potential therapeutic strategy for AD. Methods:Our coworkers previously established a fusion peptide fragment, ABAD-DP-6His, NT4, TAT, by fusing a 6-His protein to the 5 end of ABAD-DP cDNA in the adeno-associated viral vector (AAV), constructed rAAV/NT4-TAT-6His-ABADDP (rAAV/NTA). We detected the place navigation task and spatial probe test by Morris Water Maze(MWM) in the way of intranasal administration. And tested the APP and Ab1-42 expression by immunolhistochemical method; In the 7, 9, 11 months of mice age, we tested the senile plaques(SP) and apotosis expression by congon red staining and hematoxylin-eosin staining (HE staining). Results: 1. There is no statistical differences among gropes (P>0.05), in the 5 or 7 months of mice age; but in the 9, 11 months, prolonged the escape latency of place navigation test in APP grope compare with normal control grope (P<0.05&P<0.01). In the 9, 11 months of mice age, reduced the frequency of APP grope compare with the control grope in spatial probe test (P<0.05&P<0.01), increased the frequency of rAAV/ NTA grope compare with the APP grope in spatial probe test (P<0.01&P<0.01). 2. In the 5 months of mice age, there is the significant difference of APP positive expression among gropes, the APP positive expression is markable increased in APP695 mice than the C57 mice; In the 5, 7, 9, 11 months of mice age, the Ab1-42 positive expression of APP grope are more than control grope and rAAV/NTA grope; In the 5, 7, 9, 11 months of mice age, the SP and apoptosis cells are increased in APP grope compare with the control grope and rAAV/NTA grope. Conclusions:The recombinant adeno-associated virus system fusing with ABAD-DP could protect the behavior and pathology changes in AD.
