The origin of organelles (mitochondrion, chloroplast and nucleus) remainsenigmatic. The endosymbiotic hypothesis that chloroplasts, mitochondria andnuclei descend from the endosymbiotic cyanobacterium, bacterium andarchaebacterium respectively is dominant yet uncompelling, while our discoveryof de novo organelle biogenesis in the cyanobacterium TDX16 that had acquiredthe genome of its green algal host Haematococcus pluvialis overturns thishypothesis. In light of organelle biogenesis in the cyanobacterium TDX16 incombination with the relevant cellular and molecular evidence, we proposegenome hybridization hypothesis (GHH) that the origin of organelles and originof eukaryotes as well as the diversification of organelles and speciation ofeukaryotes are unified and achieved by genome hybridization: the endosymbioticcyanobacteria or bacteria obtain genomes of their archaebacterial or eukaryotichosts and hybridize with their own ones resulting in expanded genomescontaining a mixture of hybrid prokaryotic genes and eukaryotic genes, and thusthe cyanobacteria or bacteria have to compartmentalize to accommodate differentgenes for specialized function of photosynthesis (chloroplast), respiration(mitochondrion) and DNA preservation (nucleus), and consequently turn intophotosynthetic or heterotrophic eukaryotes. Accordingly, eukaryotes and theirorganelles are of multiple origin, while the formation of cancer cells is thespeciation of eukaryotes as cancer cells are new species of unicellulareukaryotes arising from bacteria. Therefore, GHH provides a theoreticalframework unifying evolutionary biology, cancer biology and cell biology anddirecting the integrated multidisciplinary research.
