As investigações sobre fungos bioluminescentes no Brasil iniciaram-se em 2001, no Laboratório de Bioluminescência de Fungos do Instituto de Química da Universidade de São Paulo. Desde então, foram identificadas 13 espécies nacionais (14% do total no mundo) pelo grupo. O fungo Gerronema viridilucens foi inicialmente utilizado como organismo modelo nos estudos mecanísticos de emissão de luz e ensaios de toxicidade. Atualmente, este fungo foi substituído pelo Neonothopanus gardneri, cujas características de crescimento e bioluminescência mostraram-se mais adequadas às linhas de pesquisa desenvolvidas. Ensaios bioluminescentes de toxicidade com fungos são importantes para avaliar a biodisponibilidade e os efeitos causados por poluentes ambientais e fungicidas. Entretanto, antes de desenvolver tais ensaios é necessário a otimização das condições de cultura, visando obter um perfil de bioluminescência mais constante e reprodutível. Este trabalho apresenta o histórico sobre fungos bioluminescentes no Brasil sob o ponto de vista do grupo. Adicionalmente, as condições de cultura foram variadas e avaliadas com o intuito desenvolver um ensaio toxicológico com o N. gardneri. Para isso, foram variados os fatores temperatura, pH do meio e concentração de nutrientes. A condição mais adequada à aplicação prevista foi definida como pH 6, não tamponado, 30ºC, 1% de melaço de cana e 0,01% de extrato de levedura. Nesta condição a emissão de luz é alta e apresenta um perfil com menos variação. Esta espécie é uma boa opção para ensaios de toxicidade, apresentando vantagens em relação ao fungo G. viridilucens. DOI: 10.5935/1984-6835.20150003The investigations on fungal bioluminescence initiated in 2001 in Brazil at the Laboratory of Fungal Bioluminescence of Instituto de Química – Universidade de São Paulo. Ever since, 13 Brazilian species (14% of total) were identified by the group. The fungus Gerronema viridilucens was used initially as model organism in the mechanistic studies on light emission and on the toxicological assay. Nowadays, this fungus was replaced Neonothopanus gardneri, whose characteristics of growth and bioluminescence are more suitable to the research conduct in the laboratory. Toxicological luminescent assays with fungi are important to evaluate the bioavailability and the effects caused by environment pollutants and fungicides. However, before the development of such assays it is necessary to optimize the culture conditions, aiming to obtain a more constant and reproducible bioluminescence profile. In this work, the history of bioluminescent fungi in Brazil is present from the point of view of the group. Moreover, the variation of culture conditions was evaluated in order to develop a toxicological assay with N. gardneri. Therefore, conditions such as temperature, medium pH and nutrients concentration were altered. The most adequate condition to the expected application was defined as pH 6, 30°C, 1% of sugarcane molasses and 0,01% yeast extract. In this condition the light emission is high and presents a low variation pattern. This species is a good option to perform toxicological assays, demonstrating benefits when compared to the G. viridilucens fungus. DOI: 10.5935/1984-6835.2015000
