近年,生物石灰化機構を調べる研究において,生物の殻と生育している環境水中の炭酸系とのつながりが注目されるよ うになり,炭酸系を継続的に分析する必要性が高まってきた.試水中の炭酸系を把握するためには,pHとアルカリ度とを 分析するのがもっとも簡便である.これは飼育実験のように採水量がごく少量に限られる試水に適用するにも都合が良い. 本報告では,海洋化学の分野で用いられているアルカリ度の一点法をもとに,試水量を1 mLにまで減らした分析手法を提 案する.重量測定により量り取り量を補正すると,アルカリ度の繰り返し精度は相対標準偏差で0.1~0.2%となった.重 量補正なしでは相対標準偏差で0.1~1.0%となった.参照海水を常に試水間に挟み分析することにより,日々の値の系統 誤差は補正できることがわかった.この手法を用いれば,pHメータのみを用いて,微量な試水中の炭酸系を生物石灰化メ カニズムの解明に十分な精度で継続的に知ることができる.
