Mesenchymal stromal cells (MSCs) have the potential to treat various disease indications and are the future of cell therapy-based regenerative medicine. Most cell-based therapies demand the availability of viable and functional cryopreserved MSCs at the treatment site. The technique of cryopreservation has its own challenge and is largely determined by the composition of various factors in the cryopreservation medium. One of the main concern of the use of traditional cryopreservation medium is the presence of xenogeneic components like serum that needs to be removed before the infusion of the final cellular product. This is a cumbersome process and requires a clean cabinet and other equipment which is not commonly available in the operation room. The immediate need is to identify a serum-free, xeno-free cryopreservation medium that can be readily used without any manipulation at the site.  We have evaluated two commercially available freezing media- CryoStor 5 (CS5) and CryoStor 10 (CS10) and compared them to the in-house PlasmaLyte A (PLA) based cryopreservation media. We analyzed post-thaw recovery, viability, phenotypic markers expression, CFU-F ability, trilineage differentiation potential and apoptosis of the MSCs cryopreserved these cryopreservation formulations at different freezing densities over a period of 6 months of cryopreservation
