古細菌の膜脂質は他の生物の膜脂質がリン脂質のエステル型であるのに対しエーテル型脂質であり, それらの生物が過酷な条件においても生育できることを可能にしている。これらの脂質はグリセロールにC20やC40の飽和イソプレン (フィタニル基やジフィタニル基) が結合している。それらの脂質は可溶性ゲラニルゲラニル二リン酸合成酵素によって合成されるか, リン酸化酵素によってゲラニルゲラニルリン酸がリン酸化されるかによってまず基質としてのゲラニルゲラニル二リン酸が生じる。これがグリセロールリン酸に結合し, 次いでプレニル基が還元されてフィタニル基になり, head-to-head縮合により二量化してジフィタニルエーテル脂質ができあがる。Sulfolobus acidocaldariusからゲラニルゲラニル二リン酸合成酵素の遺伝子を筆者らの考案したred-whiteスクリーニング系を用いてクローニングした。この遺伝子を用いて種々の組み換え体を作製して性質を調べたり, 他の同族の遺伝子と比較することによって酵素の進化についても議論ができるようになった。
