It has been reported that many methods of esterification1) or transesterification2) of various substrates with enzymes. There, water is usually used to dissolve substrates because it keeps enzymes at work whereas organic solvents do not. In case of the substrate that is not dissolved in water, an organic solvent is used but the process should be manipulated to save the performance of enzyme3). Many methods for that have been developed; a method for this is to form emulsion or reverse micelle phase in order to proceed the reaction4), and another is to apply chemical modification on the enzyme to enable function5) or use immobilized enzyme6). Recently Klibanov and his coworkers discovered that such tedious process could be eliminated. They found that the lipase functions as catalyst even in organic solvent7). Their work, however, is limited to simple molecules. We found that more complex biochemicals such as cholesterol could be esterified in a high yield and also that the recovered lipase worked more actively in repeated uses than in the first use in some organic solvents.
