However, when we attempted to deliverthe peptide drugs containing the disulfide bond, such asSomatostatin, to the brain, we faced unexpected difficulty inintroducing the dihydropyridine carrier to the peptide drugs,since the disulfide bond in the peptide structure that fixes itstertiary structure is prone to reduce into the correspondingthiol under general reduction conditions. Although thereduction of the N-methyl-pyridinium salt into N-methyl-1,4-dihydropyridine has been well described, interestingly,there are no reports on the selective reduction of the N-methylpyridinium ion into the corresponding dihydropyri-dine without cleavage of the disulfide bond. In order toachieve the brain-targeted delivery of peptides that containthe disulfide bond in their structure, we proposed to find theselective reducing agent and the reaction condition that onlyreduces the trigonellyl group in the presence of the disulfidegroup by investigating various reducing agents.
