N-Linked glycosylation is one of the most common protein modifications and can have profound effects on protein expression and function. These effects often depend on the number and position of N-linked oligosaccharides added to a protein during core-glycosylation. While the sequon, Asn-X-Ser/ Thr, serves as a recognition signal for core-glycosylation, other protein signals must also control this process, since many AsnX-Ser/Thr sequons are glycosylated inefficiently or are not glycosylated at all. A variety of experimental approaches have been used to define the signals which control core-glycosylation at specific Asn residues. These include comparisons of protein sequences near glycosylated and non-glycosylated sequons, use of sequon-containing peptides as oligosaccharide acceptors or inhibitors, and analysis of the glycosylation of sequons in recombinant proteins. Such studies reveal that a complex set of factors determines the efficiency of oligosaccharide addition at each Asn-X-Ser/Thr sequon. Amino acids near the Asn residue in a sequon can profoundly affect its oligosaccharide acceptor activity. A variety of other factors further modulate core-glycosylation efficiency by influencing the accessibility of sequons for glycosylation at a critical time during protein synthesis. Studies addressing these issues are reviewed. A. Introduction One of the most common types of eukaryotic protein modification is N-linked glycosylation, in which oligosaccharides are added to specific asparagine (Asn) residues (1,2). Nlinked oligosaccharides (N-glycans) play a critical role in the expression of most cell-surface and secreted proteins in eukaryotes by promoting their transport to the cell surface or extracellular space (3). Alteration in the number or position of N-glycans on a protein can influence its expression by altering its folding, stability, or potential to aggregate in the endoplasmic reticulum (ER) or Golgi apparatus (3, 4). The pre-synthesized core-oligosaccharide, Glc3Man9GIcNAc2, is transferred from the dolichol-P-P-oligosaccharide donor to an Asn-X-Ser/Thr sequon in a nascent protein by the enzyme, oligosaccharyltransferase. The number and position of N-glycans added to a protein are determined in the first step of N-linked glycosylation, core-glycosylation. In this step, the pre-synthesized core-oligosaccharide, Glc3Man9GlcNAc2, is transferred from the lipid donor, dolichol, to a protein by the enzyme, oligosaccharyltransferase (1, 2). Oligosaccharyltransferase and the dolicholP-P-oligosaccharide donor are integral to the ER membrane, and the active site of the enzyme resides near the ER membrane on the lumenal side (9,10). Core-glycosylation usually occurs co-translationally, as the glycosylation site on a nascent protein enters the ER lumen (Fig. 1)(11-13). In some cases, particularly in yeast systems, core-glycosylation can also occur posttranslationally (14-17). As newly synthesized proteins pass through the ER and Golgi apparatus, core-oligosaccharides are modified by glycosidase and glycosyltransferase enzymes (1, 2). This processing can generate a heterogeneous array of Nglycan structures, even at a single site in a glycoprotein. Despite the importance of N-linked glycosylation, the protein signals which control oligosaccharide addition at specific Asn residues are not well-understood. In 1974, Marshall identified the tripeptide sequence or "sequon", asparagine-Xserine or asparagine-X-threonine, (Asn-X-Ser/Thr) as the recognition signal for N-linked glycosylation (18). Since then, only rare examples of N-linked glycosylation at other sequences (e. g. Asn-X-Cys and Asn-Gly-Gly-Thr) have been described (19). It is well-established that the presence of Pro at the X position blocks glycosylation (see below). However, other protein signals must also control core-glycosylation, since many sequons in proteins are not glycosylated for reasons which are not understood (19-21).
