s TGF-β1 is the prototype of the so called TGF-βgrowth factor family. The family consists of an increasing number of different polypeptide modulators of cell growth, differentiation and morphogenesis. Numerous ways to regulate the expression of the TGF-β genes have been identified. Their expression can be affected by autocrine and paracrine mechanisms, and also by some other growth factors. TGF-β:s are also subject to regulation by retinoids, steroid hormones and vitamin D. Active TGFβ regulates transcription, and there is an increasing list of genes that are affected by TGF-β. A characteristic feature in the biology of TGF-β:s is that they are usually secreted from cells in latent forms. TGF-β was originally isolated from the conditioned medium of sarcoma virus transformed cells. The purification involved acidification, which, on the basis of current understanding, activates latent forms. Platelets and placenta are rich sources of TGF-β. We have recently found that TGF-β is notjust secreted from cultured cells but is deposited in a latent form to the pericellular space, namely to the extracellular matrix. The latent complex consists of the small latent complex (TGF-β and its propeptide)and a high molecular weight protease resistant binding protein, LTBP(latent TGF-β binding protein). Protease-mediated release from the matrix is supposedly an initial step in the activation of the molecule, which possibly occurs in a coordinate way at the cell surface. TGF-β has also a role in the regulation of wound repair, extracellular matrix formation and immunological processes. TGF-β is known as a potent chemoattractant for fibroblasts, monocytes, neutrophils and eosinophils(3-6). These effects may be beneficial in wound healing and regulation of scar formation, as well as in cell invasion processes. The background to the modulation is explained by the fact that TGF-β has a major role in the regulation of extracellular matrix synthesis and degradation. TGF-β:s have an important role in the control of the proteolytic balance of cells and in the regulation of the production and structure of the components of the extracellular matrix(for review, see 1,2,7,8). TGF-β stimulates the synthesis of various components of the extracellular matrix like collagens, fibronectin, vitronectin, tenascin and proteoglycans(9-12). It also suppresses matrix degradation by decreasing the expression of proteinases, such as plasminogen activators(13,14), fibroblast collagenase(15)and stromelysin(16), and by inducing proteinase inhibitors, such as plasminogen activator inhibifor-1(PAI-1)(13,17)and tissue inhibitor of metalloproteinase1(TIMP-1)(15). In addition, TGF-β regulates the expression of matrix receptors, the integrins(see 18). In cell culture TGF-β is secreted into the medium in an inactive form. Soon after its discovery it was understood that a peculiar feature in the biology of TGF-β:s is the fact that they are secreted from the cells in forms that needed activation to exert their effects(c.f.2,19). TGF-β was originally isolated from the medium of cultured cells, but it was later found that also platelets, placenta and bone are rich sources of it(1,2,7,8, 19,20).The isolation from tissues involves extraction by acidethanol, which is known to dissociate latent forms of TGF-β and yield biologically active molecule. Latent TGF-β isolated from cultured cells can be activated by extremes of pH, certain glycosidases and by the serine protease plasmin(see below). In studies on protease-mediated activation using plasmin, the secreted latent TGF-β from the conditioned medium of fibroblastic cells could not be completely activated(only up to 30%)while temporary acidification brought about full activation(21). Analysis of the pH dependency of TGF-β activation indicated that TGF-β may be associated with a number of different macromolecules. Plasmin is known to cleave the propeptide part of TGF-βbut not affect TGF-β itself(22). These data suggested that a fraction of TGF-β was in a form inaccessible to plasmin or that there are two different binding proteins or pools of TGF-β in the medium of cultured cells. The recent observations of the association of latent TGF-β complexes with pericellular structures and their release by proteases(23)implicate that this fraction of TGF-β may be the pool that is readily activatable by proteases. Fibronectin is an adhesive protein, which has a wide variety of binding proteins with different affinities (c.f. 24).
