The mode of action of the sulfatases in the glycosaminoglycan metabolism is briefly reviewed. The evidences so far obtained suggest that there are three pathways of degradation of glycosaminoglycans involving several specific sulfatases with distinct modes of action. In mammals, and possibly in other vertebrates, the sulfatases act on a sequential order in concert with sugar hydrolases, removing sulfates from the nonreducing ends of the molecules in each cycle of degradation. In invertebrates, the sulfatases act directly upon the polymer producing extensive desulfation without need of concomitant depolymerization which takes place only on a second stage, by action of the sugar hydrolases. Finally, in bacteria, the sulfatases act only upon sulfated disaccharides and monosaccharides formed from the glycosaminoglycans by action of eliminases and glycuronidases. The biological role of the sulfatases on the recycling of sugars and sulfate is discussed. 要 旨 グリコサ ミノグリカン代謝におけるスルファターゼの作用 様式を短くまとめた。今 までに得 られた証拠によれば、3通 り のグリコサミノグリカン分解経路が存在 し、それぞれ作用様式 の異なる、数種の特異的なスルファターゼが関与 していること が示されている。哺乳動物、おそ らく他 の脊椎動物 において も、スルファターゼは糖加水分解酵素とともに逐次的に作用 し、各分解のサイクル毎に分子の非還元末端か ら硫酸基を除去 する。無脊椎動物では、スルファターゼはポリマーに直接作用 し、ポリマーを同時に分解することな く、高度に脱硫酸化す る。脱硫酸化後、糖加水分解酵素の働きでポリマーは分解 され る。最後 に細菌のスルファターゼは、エ リミナーゼやグリクロ ニダーゼ分解 によりグリコサミノグリカンか ら生 じた、硫酸化 二糖や単糖に作用する。糖 と硫酸の再利用におけるスルファタ 。
