The hundreds of glucosphingolipids(GSLs) found in nature are derived from glucosylceramide(G1cCer). It is possible to block the synthesis of G1cCer with a synthetic analog of ceramide, PDMP(D-threo-l-phenyl-2-decanoylamino-3-morpholino-l-propanol)(Fig.1)(1,2). Thus, because of the continuous action of GSL hydrolases, PDMP acts to lower the level of every GSL. The speed of lowering depends on how completely G1cCer synthase is inhibited, on the activity of each GSL hydrolase, the susceptibility of each GSL to attack by the hydrolase(i.e., its subcellular location), and the rates of the various synthases that recycle smaller GSLs into larger GSLs.
