UDP-GalNAc : ポリペプチドN-アセチルガラクトサミン転移酵素 (GalNAc-転移酵素、GalNAc-T) はタンパク質のムチン型O-グリコシレーション (GalNAcα1-O-Ser/Thr) を触媒する酵素群である。GalNAc-T3の遺伝子変異が常染色体劣性代謝性疾患である家族性腫瘍状石灰化症 (familial tumoral calcinosis、FTC) (OMIM 211900) の原因となることが近年明らかにされた。FTCは高リン血症および異所性石灰化を伴う。我々はリン酸代謝調節因子であるFGF23の分泌にO-グリコシレーションが必要であること、およびGalNAc-T3がスブチリシン様プロプロテインコンベルターゼにより認識される配列を特異的にO-グリコシレーションし、その修飾がfurinプロテアーゼによるFGF23のプロセシングを阻害することを明らかにした。これらの結果は完全長のFGF23を産生する際のO-グリコシレーションとプロテアーゼによるプロセシングとの競合、というFGF23の新規翻訳後修飾制御モデルを示唆している。
