糖尿病領域の細胞周期異常については糖尿病性腎症 で研究が進んでおり,メサンギウム細胞では,高血糖 によりサイクリン依存性キナーゼ阻害因子である p21,p27が増加してG1期細胞周期停止が起こる 結果,細胞肥大を来たすことが知られている.一方 で,肥満における脂肪細胞の細胞周期異常に関しては, まだ十分には解明されていない.臨床上有用な糖尿 病治療薬である thiazolidinediones(TZDs)は核内受 容体作動薬のひとつであり,peroxisome proliferatoractivated receptor-γ(PPARγ)アゴニストとして作用 する.同じく核内受容体のひとつであるレチノイドX 受容体(RXR)は PPARγとヘテロダイマーを形成す るが,本研究ではRXRアンタゴニスト(HX531)を 用いて,細胞周期の観点から肥満と2型糖尿病に対す る治療効果とその分子メカニズムについて解明した.
