イヌホタルイは表1に 示す地域の水田から採取 し、秋田県大曲市の東北農業研究センターで生育 させ、株または種子で維持 した。オモダカは表2に 示す地域の水田から採取 して生育 させ塊茎で維 持 した。ALS遺 伝子については、 イヌホタルイの花茎 とオモダカの矢尻葉からmAを 抽出 し、RTPCRに よってALS遺 伝子のcDNAを 単離 し塩基配列を決定 した。イヌホタルイでは緑葉か らゲノム DNAを とりイン トロンの有無について検討 した。ALS遺 伝子の変異は、cDNAま たはゲノムDNAか ら PCRに よってALS遺 伝子を増幅 し、ダイレク トシーケンスによって解析 した。イヌホタルイの抵抗 性検定はALS活 性 を用いた迅速検定法(内 野2002)に よって行った。オモダカの除草剤反応試験 は1/5000aワ グネルポッ トに詰めた埴壌土に5月 下旬に塊茎 を植 え、処理1ヶ 月後に植物体を採取 して乾物重を調査 した。SU剤 はベンスルフロンメチル0.75%粒 剤、 ピラゾスルフロンエチル0.07% 粒剤、 チフェンスルフロンメチル75%水 和剤 を使用 し、粒剤は3葉 期に水和剤は5葉 期に処理を行 った。
