(BBTV) was not successful until 1987 when liquid nitrogen was used to freeze diseased tissue prior to pulverization and solvent extraction. The extract was subjected to overnight stirring at low temperature followed by low/high speed centrifugation19). This procedure was also successfully applied to isolates of BBTV from diseased banana in Australia5,16), Pakistan9,10) and Hawaii23). Based on available reports, BBTV is an isometric particle with a diameter ranging from 18 to 22 nm5,10,16,19). Although initially the nature of the nucleic acids associated with BBTV was confusing18), the recent consensus is that they are circular single-stranded DNAs with multiple DNAs in the genome2,6,18,22-24). With purified virus available, monoclonal and multiclonal antibodies against BBTV were subsequently obtained4,20). Monoclonal antibodies had been used to detect BBTV in different commercial cultivars7,17,21), wild banana21) and viruliferous banana aphids (Pentalonia nigronervosa Coq.)17), by means of direct enzymelinked immunosorbent assay (ELISA), and to localize BBTV in phloem cells of diseased banana leaves by indirect fluorescent antibody staining17). Since fresh tissue samples often deteriorate, during shipment to our laboratory for testing, samples may not be suitable for monoclonal antibody detection of the virus. We, therefore, developed a simple method to preserve the serological activity of BBTV in diseased tissue for longdistance shipment to diagnostic centers for testing by ELISA. Washed banana leaves (cv. Cavendish) infected with BBTV were cut into small pieces (ca. 5×5mm) and dried on the bench in the laboratory, in the sun or in an oven at various temperatures. Fresh leaf tissue stored at 4°C for the same period of time as the treatment was used as the control. The dried tissue was pulverized in a coffee grinder for 1min before the serological tests.
