The study wa s conducte d on a mode l of endothelial dysfunction cause d by intoxication wit h nickel chloride and an eNOS inhibitor – L-NAME. The result s showed a significant increas e in oxidant s and thei r inhibitory effect on the development of nitri c oxide (NO) – a biochemica l marker of the endothelia l dysfunction. When introducing nickel and modified L-arginine, the internal molecular structure of endothelia l NO synthase (eNOS) and the abilit y of the enzyme to produce nitric oxide and activ e oxygen metabolites (AMA) is violated. L-arginine stimulate s the expression o f eNOS, the development o f nitric oxide and inhibits lipoperoxidation. Thi s experimenta l method i s relevant , becaus e only fundamenta l knowledge about the mechanisms of development of endothelia l dysfunction will allow us to develop a predictive approac h for the prevention of negative effects.
