Clinical Central Laboratory, Toyama Medical an 0.5. 8, 11980. 12.1) The normalizing effects of antithrombin III and dextran sulfate on coagulation disorders of experimental disseminated intravascular coagulation (DIC) were investigated in rabbits. 1. In vitro studies : (1) Dextran sulfate and heparin at concentrations higher than 0. 3 mg/ml and 0.001 units/ml, respectively, showed anticoagulant effects resulting in prolongation of a-PTT. (2) Heparin accelerated anticoagulant activity of antit rombin III. Dextran sulfate showed moderate anticoagulant activity. 2. In vivo studies using rabbits: (1) Infusion of single reagent. Intravenous injection of antithrombin III more than 500 units or dextran sulfate more than 75 mg showed anticoagulant effects. (2) Experimental DIC. Coagulation disorders of experimental DIC induced by infusion of thromboplastin was prevented by prior intravenous administration of antithrombin III or dextran sulfate, and those induced by infusion of tissue thromboplastin were also normalized rapidly by administration of antithrombin III or dextran sulfate. Futhermore, the latter showed normalizing effects on coagulation disorders induced by infusion of human tissue thromboplastin, namely, cell lysate of acute promyelocytic leukemia and saline extract of gastric cancer tissue. Therefore, it is suggested from the above observation
