In fishes, heating conditions for the heat inactivation of complements are still indistinct. Heating conditions for the inactivation of complements were investigated using normal sera and antisera of rainbow trout Salmo gairdneri, masu salmon Oncorhynchus masou, coho salmon O. kisutch, goldfish Carassius aurantus and tilapia Sarotherodon nilotica. Antisera were prepared from these five fishes immunized with goldfish red blood cell (GFRBC) and rainbow trout red blood cell (RTRBC) as immunogens. Heating conditions for the inactivation were determined with the hemolytic system consisting of GFRBC or RTRBC and antisera heated at 30•Ž to 60•Ž for 5 to 30 min. The hemolytic activities of the antisera were assayed at 30•Ž for 60 min with hemoglobin leakage (O. D. at 541nm) from GFRBC or RTRBC. Consequently, conditions for the heat inactivation of complements were defined as follows: 42•Ž-20 min for masu salmon, 44•Ž20 min for rainbow trout, 45•Ž-20 min for coho salmon and goldfish, and 47•Ž-20 min for tilapia. Immune hemolysis reaction was achieved without trouble for the five fishes by the use of the inactivated antisera as hemolysins and the normal sera as complements.
