骨吸収因子の刺激により骨芽細胞/ストローマ細胞膜上に発現する破骨細胞分化誘導因子 (ODF) は, 破骨細胞前駆細胞膜上に発現するODF受容体であるReceptor activator of NF-κB (RANK) と結合することにより, 破骨細胞への分化を誘導する。そこで, ヒト前骨髄性白血病細胞 (HL 60) のRANK遺伝子発現ならびに破骨細胞への分化に対するIL-11の効果について検討した。HL 60は恒常的にRANK mRNAを発現しており, 骨吸収因子であるIL-11, PGE2ならびにVD3はHL 60のRANK mRNA量を濃度依存的に増加させた。またHL 60をマウス骨髄由来ストローマ細胞 (ST 2) と共存培養する際, IL-11を添加したところ破骨細胞が形成された。M-CSF単独刺激においても同様の結果が得られた。M-CSF存在下にIL-11処理を行ってもHL 60におけるRANKの発現量はIL-11単独刺激で得られた値を超えることはなかった。一方, 破骨細胞形成能はM-CSFとの同時処理により増強された。これらの結果からIL-11は, 破骨細胞前駆細胞のRANK発現を誘導することにより骨吸収の調節に関与することが示唆された。
