A study was conducted to examine the mechanism of the bactericidal effect of He-Ne laser irradiation on Streptococcus mutans, and to clarify the conditions under which the bactericidal effect was expressed. Two milliliters of 0.5% agar plus 0.5 ml of bacterial cell suspension containing crystal violet (CV) were poured onto Brain Heart Infusion agar, and the surface of the agar plates was vertically irradiated with a He-Ne laser (Acusoft, 10 mW, 632.8 nm, Osada Electric Co., Tokyo, Japan) . After laser irradiation, the plates were cultured anaerobically and also aerobically. Similarly, 0.13 ml of cell suspension containing CV (final concentration: 3.2 , ug/ml) was placed in each well of a microtitration plate, and the laser beam was used to vertically irradiate the cell suspensions.
