Fluorescent antibodies against fast skeletal, slow skeletal and cardiac myosin heavy chains (MHC) were applied to cultured cells from the chicken embryonic pectoralis muscle. Some of the spindle-shaped mononucleated cells and all the multinucleated myotubes were stained with both anti-fast and anti-cardiac MHC antibodies. In contrast, only myotubes at later culture stages were stained with anti-slow MHC antibody. The observations indicate the presence of myosin antigens in the myotubes which cross-react with MHC from fast, cardiac and to lesser degree slow muscles of the adult chicken.
