神経細胞は1本の長い軸索と複数の樹状突起を形成し, 高度に発達した細胞極性をもつ. 極性化の過程において, 神経細胞は複数の未成熟な突起をもつ対称な状態から, 急激に1本の長い突起を伸ばして対称性を破る. この長い 突起はその後も伸長を続けて軸索へと分化し, 一方, 残り の突起が樹状突起になる . 極性化の重要なステップであ る軸索形成の分子機構は, これまでのところ十分に理解さ れていない. また, 形成された軸索がいかにして正しい標 的細胞にガイダンスされるのか, 細胞内における分子のメ カニクスも良く解っていない. 本稿では, 我々の研究室で 行われてきた二次元電気泳動法を用いたプロテオーム解 析の高感度化について解説する. また, それによって同 定された Shootin1 と Singar1 の機能解析, さらに Singar1 と 相互作用する分子として同定された Rab33a の機能につい て概説する.
