It was found unexpectedly that when ethanol was applied for the purpose of anaesthesia of frog sartorius muscles, the amplitude of the end-plate potentials (e. p. p. s) recorded extracellularly was increased and the changes in the amplitude in various kinds of bathing solutions containing ethanol were shown previously (OKADA and ADACHI, 1961). It has been reported that methanol, ethanol and acetone have an anticholinesterase action on the skeletal neuromuscular system (FENG and LI, 1941) and that ethanol facilitates the synaptic transmission of stelate ganglia of cat (LARRABEE and POSTERNAK, 1952). It has also been shown recently that ethanol, methanol and n-propanol act on the neuromuscular junction of diaphragm of cat preand post-synaptically, by observing e. p. p. s and miniature end-plate potentials (m. e. p. p.s) (GAGE, 1965). After introduction of the microapplicating method of acetylcholine on the end-plate (NASTUK, 1953), the function of the nerve terminal of the neuromuscular junction and of the end-plate receptor membrane has almost been clarified (DEL CASTILLO and KATZ, 1955, 1956a, b; KOKETSU, 1958; DUDEL, 1965) and several hypotheses on the mechanisms of the neuromuscular chemical transmission have been proposed (FATT and KATZ, 1952a, b; THESLEFF, 1960; OKADA, 1961; KOELLE, 1962). In the present experiments, the changes in e. p. p. s, m. e. p. p. s and acetylcholine potentials (Ach potentials) produced by several kinds of alcohols and acetone were investigated intracellularly and it was found that the effect of alcohols on the frequency of m. e. p. p. s was especially dramatic. The results obtained will be discussed on the mechanism of action of alcohols and acetone pre-and post-synaptically.
