Plants synthesize substances, such as phenolic compounds, as a defense to adverse environmental conditions. In this context, the addition of contaminants to the culture medium can induce specific stress responses in some plant organisms. The present study evaluated in vitro development of R. graveolens in culture medium artificially contaminated with benzo[a]pyrene and phenanthrene, as well as the production of total flavonoids in shoots grown under these conditions. Eight months old sprouts maintained in vitro on Murashige/Skoog medium without growth regulators were used. The solidified media were supplemented with phenanthrene (1.0; 5.0 and 10.0 mg/L) or benzo[a]pyrene (0.001; 0.010 and 0.100 mg/L) and analyzes were performed over 21 days of culture. The total phenolic compounds were determined by the Folin-Ciocalteau method, while the quantification of different flavonoid fractions was performed using the colorimetric technique adapted from Maia and Park. Species showed capacity for in vitro development and tolerance to the presence of hydrocarbons tested, thus revealing the potential for cultivation in areas contaminated by these compounds at the levels investigated. Analysis of phenolic compounds and total flavonoids indicated that shoots maintained under these conditions can develop at the maximum concentrations tested (10 mg / L of phenanthrene and at least 0.1 mg / L of benzo[a]pyrene), without changing their phytochemical profile about rutine.
