【目 的 】 これ までに我 々は,定 常状態においてキノロ ン系抗菌剤の脳組織 濃度及 び脳脊髄液(CSF)濃 度 が 血清中非結合型濃度 と比較 して低 く保 たれているこ とを明 らか として きた1).そ こで,本 研究では, これ ら薬剤 の中枢 への移行性 を制限 している要因 を,中 枢神経系 での薬物分布 を記述 したモデル2), dis面butedm(⊃de1(Fig.1)を 用 いて定量的に評価す る ことを目的 とした.種 々のキノロン系抗菌剤の間 でそ の中枢 移行 性が異 なる原 因 を明 らか とするた め,6種 の化合物,nornoxacin(NFLX),AM-1155, ofloxacin(OFLX),fleroxacin(FLRX),sparfloxacin (SPFX)及 びpefloxacin(PFLX)の ラッ トにおける中枢 動態 を比較,検 討 した.
