Electron microscopic studies on the bone marrow cells in 15 cases of Ig-G myeloma and 3 cases of Ig-A myeloma and 3 cases of Waldenstrom's macroglobulinemia were carried out.In one case of macrogrobulinemia the plasma cell was predominant in the bone marrow, but it was not so in the other two cases. Therefore, to elucidate the macroglubulin-synthesizing cell, ferritin antibody technique was applied to bone marrow cells of these 2 cases.1. Uniformly proliferated myeloma cells were morphologically only slightly different from case to case, except for one case of macroglobulinemia where several stages of maturity of the plasma cell were found. In comparison with the normal plasma cell the specific features of the myeloma cell were irregular configurations of the nucleus, occasionally dispersed chromatin, intranuclear inclusion bodies, variously-developed rough-surfaced endoplasmic reticulum, prominent Golgi area, many dense bodies and the fibrillar structure in the cytoplasm.2. Intranuclear inclusion bodies, cytoplasmic and lysosomal crystalloids and the Russell bodies were accountable for the excess production of γ-globulin in the cell.3. According to the ferritin antibody technique, ferritin particles were found in only two kinds of marrow cells: the plasma cell and the large lymphocyte with rich cytoplasm, a prominent Golgi area, numerous mitochondria and abundant free ribosomes. This finding suggests that these cells might synthesize macroglobulin.
