Platelets are activated at sites of vascular injury via several molecules, such as adenosine diphospha te, collagen and thrombin. Full platelet aggregation is absolutely e ssential for normal hemostasis. Moreover, this phys iological event can trigger circulatory disorders, such as thrombosis, atherosclerosis, and cardiovascular disease. Theref or , platelet function inhibition is a promising approach in preventing pl ate et-mediated circulatory disease. Many studies r eported the involvement of mitogen-activated protein kinases (M APKs) signaling pathways in platelet functions. How ever, these studies were limited. Thus, we examined MAPK signal ing pathways in human platelets using specific MAPK inhibitors, such as PD98059, SB203580, and SP600125. We observe d that these inhibitors were involved in calcium mo bilization and influx in human platelets. They also suppressed thrombin-induced αand δ-granule release. These results suggest that PD98059, SB203580, and SP600125 exhibit Ca 2+ antagonistic effects.
