В процессе хранения эритроциты подвержены риску активации свободно-радикальных процессов и перекисного окисления липидов (ПОЛ) их мембран, вследствие чего могут изменяться структурно-функциональные свойства клеток. Цель работы – определение уровня накопления продуктов перекисного окисления липидов в эритромассе при хранении их влияния на стойкость эритроцитов. Исследуемые показатели измеряли в образцах эритромассы доноров, хранящейся в условиях станции переливания крови при oС с гемоконсервантом «Глюгицир» («Биофарма», Украина): сразу после забора крови (контроль) и после хранения в течение , и -х недель. Предварительно суспензию эритроцитов отмывали трижды с целью освобождения от следов гемолиза. Эритрограммы кислотного гемолиза ( , н HCl) для определения стойкости эритроцитов получали с помощью агрегометра-формометра [S.V. Rudenko, ]. Концентрации первичных (диеновых (ДК), триеновых, оксодиеновых и тетраеновых конъюгатов) и вторичных продуктов ПОЛ определяли спектрофотометрически. При расчете концентраций первичных продуктов ПОЛ использовали коэффициенты молярных экстинкций [В.Б. Гаврилов, М.И. Мишкорудная, ; Ф.Д. Ганстон, ]. Показатели кислотной резистентности при хранении эритромассы изменялись волнообразно. После недельного хранения значимо снижалась устойчивость клеток к гемолитику, к концу второй недели вид эритрограмм возвращался к контрольному, а после трех недель хранения эритрограммы представляли среднее между графиком контроля и графиком первой недели хранения. После трехнедельного хранения устойчивость всех субпопуляций эритроцитов снижалась. Показатели перекисного окисления липидов повторяли тенденцию изменения эритрограмм – самые высокие цифры после первой недели хранения (ДК в контроле – ( , ± , ), относительно первой недели ( , ± , ) нмоль/мл). После двухи трехнедельного хранения, вероятно, постепенно истощается пул субстратов ПОЛ, а также происходят метаболические изменения в эритроцитах (ДК – ( , ± , ) и ( , ± , ) нмоль/мл соответственно). Таким образом, в процессе хранения эритроцитов изменяются их структурно-функциональные свойства, происходит метаболическая перестройка. При этом особому влиянию подвержен пул молодых эритроцитов, имеющих большее количество полиненасыщенных жирных кислот в мембранах, чем пул старых. During storage the erythrocytes are exposed to the risk of the free-radical and lipid peroxidation (LPO) process activation of their membranes, which may possibly entail the changes in structural and functional properties of cells. The research aim was to determine the level of lipid peroxidation product accumulation in RBCs during storage and its impact on erythrocyte resistance. The studied indices were measured in the donor RBCs specimens, stored under the blood supply service at 4oС with blood preservative Glugicir (Biopharma, Ukraine), i. e. right after blood sampling (the control) and after 1-, 2and 3-week storage. The erythrocyte suspension was preliminarily washed thrice to remove the hemolysis traces. The erythrogramms of acid hemolysis (0.1 n HCl) to determine the erythrocytes resistance were obtained with an aggregometer [S.V. Rudenko, 1998]. The concentrations of primary (conjugated dienes (CD), trienes, oxidienes, tetraenes) and secondary LPO products were determined spectrophotometrically. When estimating the concentrations of primary LPO products the molar extinction coeffi cients were used [V.B. Gavrilov, M.I. Mishkorudnaya, 1983; F.D. Gunston, 1986]. Indices of acid resistance during RBCs storage were wave-like changed. After the 1-week storage the cell gemolytic resistance signifi cantly decreased, by the end of the 2-week one the erythrogramm returned to the control, after 3 weeks the erythrogramms represented the mean between the control graphs and that for 1-week storage. After 3-week storage the resistance of all the erythrocyte subpopulations decreased. The LPO indices repeated the tendency of erythrogramm change, i. e. the highest numbers were after 1-week storage (CD in control were (46.65 ± 4.38) vs 1-week one (288.61 ± 53.79) nmol/ml). After 2and 3-week storage, the pool of LPO substrates was probably depleted, as well as the metabolic changes in erythrocytes occurred (CD indices were (17.58 ± 1.25) and (32.68 ± 4.48) nmol/ml, respectively). Thus, during erythrocyte storage their structural and functional properties changed and metabolic rearrangements occurred. Herewith, the pool of young erythrocytes with a greater amount of polyunsaturated fatty acids was mostly aff ected than that of old ones.
