Концентрати еритроцитів, отриманих від здорових донорів, представлені клітинами різного віку ( – діб), співвідношення яких у крові може змінюватися залежно від віку та статі донора. У процесі зберігання старі клітини більшою мірою піддаються пошкодженню, ніж молоді. Розмір старих еритроцитів внаслідок втрати клітинної мембрани та води стає меншим, ніж молодих. У випадку підвищення внутрішньоклітинної концентрації гемоглобіну змінюються кислород-транспортна функція, осмотична крихкість, ригідність тощо. Концентрати еритроцитів трьох здорових донорів були розділені у градієнті щільності Percoll та відібрані за фракціями: молоді (менш щільні) та старі (більш щільні). Клітини зберігалися протягом діб при °C. Кожні діб визначали динаміку змін індексів еритроцитів, рівень гемолізу, насичення киснем, осмотичну крихкість, здатність до деформації, ригідність та екстерналізацію фосфатидилсирину. Вплив тривалості зберігання на якість еритроцитів оцінювали за допомогою повторного аналізу дисперсій ANOVA (p < 0,05). Молоді еритроцити після розділення показали значуще вищий середній корпускулярний об’єм (СКО) і нижчу середню концентрацію корпускулярного гемоглобіну (СККГ) порівняно з контрольними клітинами у концентраті і старими еритроцитами (р < , ) після -добового зберігання. Старі еритроцити мали значуще скоріше збільшення СКО та значуще зменшення СККГ (р < , ). Аналіз зберігання показав значуще меншу здатність до переносу кисню старих еритроцитів порівняно з контрольними (р = , ) та молодими (р < , ) еритроцитами, але швидкість змін була однаковою в обох субпопуляціях. Деформованість, осмотична крихкість і екстерналізація фосфатидилсерину молодих і старих еритроцитів не відрізнялися від контрольних клітин, тоді як швидкість змін ригідності і осмотичної крихкості була значно вищою в обох субпопуляціях еритроцитів. У процесі зберігання молоді клітини мали більш низьку ригідність, ніж несепаровані (р = , ) та старі (р = , ), але даний показник змінювався скоріше у процесі зберігання (р = , ), як й осмотична крихкість (р < , ). Рівень гемолізу у субпопуляціях старих еритроцитів був значуще вищим (р = , ) і швидше зростав (р = , порівняно з молодими, проте обидві популяції мали вищий рівень гемолізу, ніж несепаровані клітини (р < , ). Таким чином, відмінності якісних показників молодих та старих субпопуляцій еритроцитів після -добового зберігання свідчать про значний внесок у пошкодження концентратів крові, які мають більш високий вміст старих клітин. Red cell concentrates (RCCs) received from healthy blood donors contain an age spectrum of cells (from 0 to 120 days old) that can vary depending on donor’s age and sex. Older cells may contribute to the red blood cells (RBCs) storage lesion to a greater extent than younger cells. Due to progressive loss of cell membrane and water with age, older RBCs become smaller than younger RBCs and their intracellular hemoglobin concentration is higher, resulting in changes in RBC characteristics such as oxygen carrying capacity, osmotic fragility, rigidity, and others. RCCs from three healthy donors were separated with Percoll density gradients into less dense (young) and dense (old) RBCs and stored for 42 days at 4°C. In vitro quality was assessed weekly to monitor changes in RBC indices, hemolysis, oxygen saturation, osmotic fragility, deformability, rigidity and phosphatidylserine externalization (Annexin V). Repeated measures analysis of variance (ANOVA) was used to determine the infl uence of storage duration. Signifi cant diff erence was defi ned as p < 0.05. Young RBCs after Percoll separation showed signifi cantly higher mean corpuscular volume (MCV) and lower mean corpuscular hemoglobin concentration (MCHC) compared to both control cells (parent RCCs) and old RBCs (р < 0.001) throughout the 42-day storage. Old RBCs had a signifi cantly faster increase in MCV and decrease of MCHC comparatively (р < 0.001). Analysis of the storage eff ect showed signifi cantly reduced oxygen carrying capacity in old RBCs compared to the control (р = 0.002) and young RBCs (р < 0.001) throughout storage, but the rate of changes were the same for both subpopulations. Deformability, osmotic fragility and phosphatidylserine externalization of young and old RBCs did not differ from the control cells, while the rate of the change in rigidity and osmotic fragility were signifi cantly higher in both RBC subpopulations. Young RBCs showed lower rigidity than control (р = 0.003) and old ones (р = 0.0014), but a higher rate of change for both rigidity (р = 0.003) and osmotic fragility (р < 0.001). Hemolysis in the old RBC subpopulation was signifi cantly higher (р = 0.048) and increased rapidly (р = 0.009) in comparison with the young cells during storage. However, both the separated RBC subpopulations had a higher percent of hemolysis compared to control cells during the storage (р < . ). Therefore, the diff erences in the quality parameters of the young and old RBC subpopulations during 42 days of storage suggest increased contribution to the storage lesion in blood samples containing a higher ratio of old RBCs.
