Область температур Tg+ΔT (Tg – температура расстеклования аморфных фракций в замороженных водных растворах криопротекторных веществ, ΔT– интервал температур в пределах ... °С) является сложной для экспериментального изучения описывающих ее диаграмм состояния. Традиционные для таких исследований методы дифференциального термического анализа и дифференциальной сканирующей калориметрии не позволяют четко разделить процессы обычной кристаллизации и кластерной, протекающей вблизи Tg без разрыва водородных связей между молекулами воды и криопротекторного вещества. При значениях меньше некоторой концентрации Cg(Cg – %) такое разделение фиксирует объемная сканирующая тензодилатометрия [А.И. Осецкий и др., ]. Этот факт связан с тем, что при концентрациях криопротекторного вещества Cβ, удовлетворяю щих условию Cβ < Cg, образование кластеров происходит на основе микрокристаллов льда aS и сопровождается ярко выраженными объемными эффектами. Однако при Cβ > Cg эффективность этого метода исследований снижается. Таким образом, с точки зрения теории фазовых превращений интервалы температур Tg + ΔT и концентраций Cg ΔCβ, где ΔCβ находятся в пределах – масс.%, остаются недостаточно изученными. В связи с этим была разработана новая экспериментальная методика изучения диаграмм состояния на основе термопластического анализа замороженных криопротекторных растворов. Показано, что экспериментально полученные термопластические кривые (T) водных растворов ДМСО, глицерина, ПЭОпозволяют установить природу кластерной кристаллизации в указанных температурных и концентрационных интервалах и определить ее термодинамические параметры. Эти параметры позволяют построить полные диаграммы состояний исследуемых криопротекторных растворов. При этом впервые подтверждено, что при Cβ > Cg в водных растворах ДМСО и ПЭОкластеры могут образовываться на основе микрокристаллов криопротекторного вещества. В результате сформулирован принцип построения полных диаграмм состояния криопротекторных растворов в диапазоне концентраций C ...C и диапазоне температур от Tm (температуры плавления чистых компонентов) до Tg.Также установлено, при используемых в криобиологии режимах замораживания необходимо разделять диаграммы, описывающие этапы охлаждения и последующего отогрева, ввиду их различия. Эти особенности необходимо учитывать при оптимизации технологий криоконсервирования биологических систем. The temperature range Tg+ΔT (Tg is the devitrification temperature of amorphous fractions in frozen aqueous solutions of cryoprotective substances, and ΔT is the temperature range within 1...50°C) is diffi cult for experimental study of the state diagrams, which describe it. The methods of diff erential thermal analysis (DTA) and diff erential scanning calorimetry (DSC), which are traditional for such studies, do not clearly distinguish between ordinary crystallization and cluster crystallization occurring near Tg without breaking the hydrogen bonds between water molecules and those of cryoprotective substance. At the values less than a certain concentration of Cg (Cg45–65%), such a separation is fi xed by the volume scanning tensodilatometry [A.I. Osetsky et al., 2016]. This is due to the fact that at the concentrations of cryoprotective substance Cβ, meeting the condition Cβ > Cg, the cluster formation occurs on the basis of aS ice microcrystals and is accompanied by pronounced volumetric eff ects. However, when Cβ >Cg, the effi ciency of this technique decreases. Thus, in terms of the theory of phase transformations, the temperature ranges Tg + ΔT and the concentrations Cg ± ΔCβ, where ΔCβ lies within the range of 10–20% by weight, have remained poorly studied. In this regard, a new experimental technique for studying the state diagrams based on thermoplastic analysis of frozen cryoprotective solutions has been developed. It was shown that the experimentally obtained thermoplastic curves (T) of aqueous solutions of DMSO, glycerin, PEO-1500 allowed to establish the nature of cluster crystallization within the mentioned temperature and concentration ranges and to determine its thermodynamic parameters. These parameters enable building the complete state diagrams of the studied cryoprotective solutions. Moreover, it has been fi rst confi rmed that when Cβ > Cg, in DMSO and PEO-1500 water solutions, the clusters may be formed on the basis of microcrystals of cryoprotectant substance. As a result, the principle of plotting the complete state diagrams of cryoprotective solutions within the range of concentrations C ...C and the range of temperatures from Tm (melting point of components) up to Tg, has been formulated. There has been also established the fact, that with the freezing regimens used in cryobiology, it is necessary to separate the diagrams describing the stages of cooling and subsequent heating because of their difference. These features should be considered when optimizing the cryopreservation technologies for biological systems.
