В системе контроля качества при производстве антирабических препаратов используют стандартный штамм вируса. Точность результатов контроля зависит от стабильности вируса при долгосрочном хранении. Цель работы – изучить изменение инфекционной активности вируса бешенства штамма CVS в процессе его хранения в защитных средах с сахарозой, глицерином и мальтозой при разных температурах в течение года. Объектом исследования был стандартный фиксированный штамм CVS. Вирус накапливали путем заражения пристеночного монослоя культуры клеток ВНК/с в поддерживающей среде на основе DMEM с добавлением , % фетальной сыворотки крупного рогатого скота. Вирус помещали на хранение в поддерживающей среде без добавок и с добавлением сахарозы ( %), глицерина ( %), сахарозы с глицерином (по %) и мальтозы ( %). Образцы хранили при , , – , – , – °С. Сохранность оценивали в течение месяцев (срок наблюдения). Установлено, что наиболее значимый вклад в изменение инфекционной активности стандартного штамма вируса при его хранении вносила температура, а затем срок хранения и состав защитной среды. Полная инактивация вируса во всех средах произошла в процессе хранения при °С в течение -х месяцев. Через месяцев хранения при °С в среде с добавлением смеси сахарозы и глицерина сохранилось % инфекционной активности вируса. Во всех остальных средах полная инактивация вируса при этой температуре произошла в течение – месяцев. В образце со смесью сахарозы и глицерина при °С вирус инактивировался между и -м месяцами хранения. В процессе хранения при – и – °С в течение месяцев наиболее высокие показатели сохранности вируса были в образцах с сахарозой, глицерином и их смесью – ( – ) и ( – )% соответственно. Через месяцев инфекционная активность вируса сохранилась на исходном уровне (до %) также в образцах с сахарозой, глицерином и их смесью при температуре – °С. Таким образом, для хранения при низких температурах вируса бешенства (штамм CVS) в качестве консервирующих сред целесообразно использовать культуральную среду DMEM с добавлением сахарозы ( %), глицерина ( %) или их смеси. Наиболее эффективен температурный режим хранения вируса в этих средах при – °С. Положительные температуры непригодны даже для кратковременного хранения изучаемого штамма вируса бешенства. In the quality control system, a standard strain of the virus is used in production of rabies biologicals. The accuracy of the control results depends on stability of the virus during long-term storage. The aim of the study was to investigate the change in the rabies virus CVS strain infectious activity during its storage in protective media with sucrose, glycerol and maltose at diff erent temperatures during the year. The research object was the standard fi xed CVS strain. The virus was accumulated by infecting the near-wall monolayer of the ВНК-21/с13 cell culture in the DMEMbased supporting medium supplemented with 0.5% fetal cattle serum. The virus was placed for storage in supporting medium without additives and with the addition of sucrose (5%), glycerol (5%), sucrose with glycerol (5% each) and maltose (5%). Samples were stored at 5, 37, –20, –80, –196°C. The preservation rate was assessed for 12 months (observation period). It was found that the most signifi cant contribution to the change in infectious activity of standard virus strain during its storage was made by temperature, and then by the storage period and composition of protective medium. During storage at 37°C, complete inactivation of the virus in all media occurred within 3 months. At 5°C after 6 months in the medium with addition of the mixture of sucrose and glycerol, 13% of virus infectious activity remained. In all other media, a complete inactivation of the virus at this temperature occurred within 3–6 months. In the samples with mixture of sucrose and glycerol at 5°C, the virus was inactivated between 6th and 12th months of storage. During storage at –20 and –80°C for 12 months, the highest virus preservation rates were in the samples with sucrose, glycerol and their mixture, i. e. (67–72) and (82–84)%, respectively. At the temperature of –196°C after 12 months, also in the samples with sucrose, glycerol and their mixture, the virus infectious activity remained at the initial level, i. e. up to 99%. Thus, for storage of rabies virus CVS strain at low temperatures, the use of DMEM culture with sucrose (5%), glycerol (5%) or the mixture of these as preservative media is advisable. The most eff ective temperature of virus storage in these media is –196°C. Positive temperatures are unsuitable even for a short-term storage of the studied strain of rabies virus.
