В останні роки значно підвищився інтерес до трансфузії крові тварин [K. Yagi, 2016]. Кріоконсервування є сучасним засобом тривалого зберігання компонентів крові. Для заморожування еритроцитів крові людини традиційно використовують кріозахисні середовища на основі гліцерину або 1,2-пропандіолу, але вони мало ефективні для еритроцитів бика, кроля, коня, котів і собак [D. Pogozhykh, 2017; O.M. Дєнісова, 2006]. Для кріоконсервування багатьох біологічних об’єктів більш ефективними є комбіновані кріозахисні середовища. В біомедичних дослідженнях для оцінки життєздатності клітин і візуалізації клітинних структур використовують флуоресцентні барвники. Мета роботи – вивчити можливість застосування флуоресцентних барвників для оцінки стану еритроцитів коня і бика після кріоконсервування в комбінованих кріозахисних середовищах. Відмиті еритроцити коня і бика змішували з кріозахисними середовищами у співвідношенні 1:1, інкубували при кімнатній температурі протягом 15 хв. Зразки заморожували зануренням у рідкий азот, відігрівали на водяній бані. Від кріоконсервуючого середовища клітини відмивали один раз 0,6 М NaCl і два рази 0,15 М NaCl, рН 7,4. Для забарвлення еритроцитів використовували флуоресцентні барвники Square-460 і 3-DAB («SETA BioMedicals», США). Флуоресцентні дослідження здійснювали за допомогою проточного цитофлуориметра «FACS Calibur» («BectonDickinson», США) і мікроскопа «AxioObserverZ1» («Carl Zeiss», Німеччина). Виявлено, що Square-460 і 3-DAB добре забарвлюють мембрани цілих еритроцитів і мають обмежену проникність в їх середину, про що свідчить менш яскраве світіння цитоплазми. При пошкодженні мембран еритроцитів спостерігається збільшення флуоресценції клітин, що пов’язане зі збільшенням кількості гідрофобних місць зв’язування, оскільки для барвників становляться доступними внутрішньоклітинні структури. За даними проточної цитофлуориметрії після заморожування-відтавання еритроцитів коня в присутності кріозахисного середовища на основі ДМСО спостерігається збільшення кількості подій у правій частині точкової діаграми на 10,5%, відмивання еритроцитів коня від кріопротектора не приводить до відновлення розподілення клітин. Після заморожування-відтаювання у присутності комбінованого середовища, що містить ДМСО і ПЕО-1500, зміни на гістограмі спостерігалися тільки для 1,7% еритроцитів коня, після відмивання від середовища розподілення клітин поверталося до контрольних показників. Таким чином, флуоресцентні барвники можна з успіхом використовувати для оцінки стану деконсервованих еритроцитів. Застосування комбінованих кріозахисних середовищ дозволяє значно покращити результати кріоконсервування еритроцитів коня і бика. Recently the interest of researchers in animal blood transfusion has significantly increased [K. Yagi, 2016]. Cryopreservation is a reliable method for long-term storage of blood components. Either glycerol or 1,2-propanediolbased cryoprotective media are traditionally used for human erythrocyte freezing, but they have low efficiency for bull, rabbit, equine, feline and canine erythrocytes [D. Pogozhykh, 2017, O.M. Denisova, 2006]. Combined cryoprotective media are more efficient for cryopreservation of many biological objects. In biomedical studies one uses fluorescent dyes to evaluate the cell viability and visualize cell structures. This research aim was to study the possibility of using fluorescent dyes to assess the state of equine and bovine erythrocytes after cryopreservation in combined cryoprotective media. The equine and bovine erythrocytes were washed, mixed with cryoprotective media in 1:1 ratio and incubated at room temperature for 15 min. The specimens were frozen by immersion into liquid nitrogen, and thawed in a water bath. The cells were washed from the cryopreservation medium once with 0.6 M NaCl and twice with 0.15 M NaCl, pH 7.4. For erythrocyte staining the fluorescent dyes Square-460 and 3-DAB (SETA BioMedicals, USA) were used. Fluorescence studies were performed using FACS Calibur flow cytometer (BD, USA) and AxioObserverZ1 microscope (Carl Zeiss, Germany). The Square-460 and 3-DAB were revealed to stain well the whole erythrocyte membranes and to have a limited permeability into the cell, as evidenced by less bright luminescence of the cytoplasm. During erythrocyte membrane damaging there was observed an enhanced fluorescence of cells, associated with an increased number of hydrophobic binding sites, since the intracellular structures were getting available for dyes. According to the flow cytometry data, after equine erythrocyte freeze-thawing in the presence of DMSObased cryoprotective medium there was observed an increased number of events by 10.5% in the right side of point chart. The equine erythrocyte washing of cryoprotectant entailed no recovery of cell distribution. After freeze-thawing in the presence of the combined DMSO and PEO-1500-contained medium, the changes in histogram were observed only for 1.7% of equine erythrocytes, and after washing of the medium the cell distribution returned to the control values. Thus, the fluorescent dyes may be successfully applied to assess the state of frozen-thawed erythrocytes. The use of the combined cryoprotective media enables to significantly improve the cryopreservation outcomes for equine and bovine erythrocytes.
