Использование низкомолекулярнй фракции (НМФ) из криогемолизированной кордовой крови человека (ККЧ) способствует восстановлению морфологических свойств эритроцитов донорской крови человека после гипотермического хранения (4...6°С) [Е.Е. Жаркова, А.К. Гулевский, 2015]. Добавление НМФ к клеткам крови, в частности лейкоцитам, способствует улучшению их функциональных свойств (реакции фагоцитирования) [О.Л. Горина, 2011 ]. Добавление ингибиторов гликолиза показало, что процесс стимулирования, по-видимому, связан с изменением метаболизма и транспортом глюкозы. Ранее [Ю.С. Ахатова, 2015] было показано, что компоненты криогемолизированной НМФ ККЧ стимулируют процессы гликолиза и гликонеогликолиза, способствуя увеличению энергетического потенциала клеток. В связи с вышеизложенным, целью нашей работы было изучение влияния НМФ ККЧ с молекулярной массой до 10 кДа на морфологические свойства эритроцитов и метаболизм важнейших для функционирвания красных кровяных клеток соединений (аденозинтрифосфат и 2,3-дифосфоглицерат) на 1, 7, 14 и 21-е сутки гипотермического хранения донорской крови человека. Эритроциты инкубировали с НМФ ККЧ до 10 кДа и препаратом сравнения «Актовегин»® («Nikomed», Австрия) после гипотермического хранения при температуре 37°С на протяжении часа, конечная концентрация фракции и препарата в образце составляла 0,6 мг/мл, после чего методом световой микроскопии проводили морфологическую оценку мазков. Определение содержания АТФ и 2,3-ДФГ рассчитывали спектрофотометрически при длине волны 660 нм [И.Л. Виноградова и соавт., 1980] (наша модификация). Эксперименты проводили на 1, 7, 14 и 21-е сутки после гипотермического хранения (4–6°С). Результаты проведенных исследований показали, что добавление НМФ ККЧ до 10 кДа, как и препарата сравнения «Актовегина», значимо стимулирует восстановление «рабочей» формы эритроцитов – нормоцитов (на 40–50% в зависимости от суток гипотермического хранения), снижает количество непереходных форм (сфероцитов) и повышает концентрацию АТФ и 2,3-ДФГ в донорской крови человека. Таким образом, НМФ ККЧ и препарат «Актовегин» обладают свойствами реабилитирующих агентов, т. е. способностью восстанавливать морфофункциональные свойства эритроцитов донорской крови человека. The low molecular fraction (LMF), derived from cryohemolyzed human cord blood (HCB) promotes the recovery of morphological properties of human donor blood erythrocytes under hypothermic storage (4...6°C) [E.E. Zharkova, A.K. Gulevsky, 2015]. The LMF supplement to blood cells, leukocytes, in particular, improves their functional properties (phagocytosis reaction) [O.L. Gorina, 2011]. The supplement of glycolysis inhibitors demonstrated this process to be apparently associated with a change in metabolism and glucose transport. The components of cryohemolyzed HCB LMF were reported [Yu.S. Akhatova, 2015] to stimulate glycolysis and glyconeoglycolysis, augmenting thereby a glycolytic potential of cells. Proceeding from the mentioned above the research aim was to study the effect of HCB LMF of molecular weight below 10 kDA on morphological properties of erythrocytes and metabolism of the compounds (ATP and 2,3-DPG), being essential in red blood cell functioning to days 1, 7, 14 and 21 of hypothermic storage of human donor blood. Erythrocytes were incubated with HCB LMF below 10 kDа and the reference drug Actovegin (Nikomed, Austria) after hypothermic storage at 37°C for an hour, with 0.6 mg/ ml final concentration of fraction and drug in the specimen. Afterwards the smears were morphologically assessed with light microscopy. The ATP and 2,3-DPG contents were determined spectrophotometrically at a wavelength of 660 nm (I.L. Vinogradova et al., 1980, our modification). The experiments were performed to days 1, 7, 14 and 21 after hypothermic storage (4–6°C). Our studies demonstrated the supplement of both HCB LMF below 10 kDa and reference drug Actovegin to significantly stimulate the recovery of the ‘normal’ population of erythrocytes, i. e. normocytes (by 40–50%, depending on day of hypothermic storage), to reduce a number of spherocytic erythrocytes, and in-crease the ATP and 2,3-DPG concentration in human donor blood. Thus, HCB LMF and Actovegin have the features of rehabilitating agents, i. e. the capability to restore morphofunctional properties of human erythrocytes.
