(20, 60mg/kg・ 体重)を1回 経 口投与 し, 2日 間絶 食 後 ネ ンブ タ ール麻 酔下 で心 採 血後 膵 臓 を摘 出 した.対 照 は,胡 麻油10ml/kg・ 体 重を 経 口投 与 した.膵 島 の単 離 は,膵 臓 を コラ ゲナ ーゼで 消 化 しFicoll-Conray非 連 続 濃 度遠 心 法 で行 った5).膵 島 の分散 細 胞 は,単 離 膵 島 を0.04% EGTAで5~6分 処 理 後 デ スパ ーゼ で 消 化 し分 離 した5).膵 島 のcAMP量 に対 す るTPTCl投 与 の影 響 は,単 離 膵 島5個/2ml(5%牛 胎 児血 清(FBS) 加DM 160培 地)を5% CO2下, 37°C 4時 間孵 置後, 1mMイ ソブ チル メチ ルキ サ ンチ ン存 在下 で フ ォルス コ リン(0, 5, 10μM)で45分 刺 激後 煮 沸水 中 で反 応 を停 止 した. cAMP量 は 膵 島を 凍 結融 解後 超 音波 破 砕 し, 遠 心後 の上 清 に つ い てcAMP RIAキ ッ ト(ヤ マサ) に よ り測定 した.細 胞 内Ca2+に 対す るTPTCl投 与 の 影響 は,単 離膵 島の分 散 細胞(105cells/dish)を カバ Fig. 1.
