Circulating progesterone levels in 10 anestrus 4 1/2-year-old ewes with silicone rubber progesterone implants were studied. 100 mg progesterone implants were surgically inserted and left in place for 10 days during which time blood samples were taken for determining plasma progesterone by radioimmunoassay. Prior to tratment the mean level was .1 ng/ml. After 24 hours it rose to .58 ng/ml (p< .01) and after 3 days it declined steadily to a baseline level. In vitro studies implants were shaken in an incubator for 10 days. The leakage rate was similar for all 10 days and in each case a saturated solution of progesterone was obtained. In vitro leakage from implants removed from experimental ewes was similar to those of other implants. It was concluded that the decrease in peripheral progesterone in ewes while the implant was retained was due to a physiological process rather than to the implants.
