Hormonal requirements for the uterine attachment reaction and blastocyst implantation in the mouse hamster and guinea pig were investigated. Each group of spayed species was prepared for fixation of their tissues 15-20 days after ovariectomy and subsequent progesterone treatment. Mice and hamsters received progesterone 2.5 mg daily and the guinea-pigs 4 mg/day. Electron microscopy studies revealed that an attachment of opposing luminal surfaces was present in the mouse and hamster after a 4-day treatment and in the guinea pig after a 6-day treatment. Closure of the uterine lumen did not follow upon progesterone administration in the rat. The rat epithelial trophoblast boundaries in pregnant spayed mice were then examined in a state of experimentally delayed implantation by progesterone. 4 mice were ovariectomized 2 days after mating and given 2.5 mg for 6 days. The results showed that although the uterine attachment reaction was present the cell membrane of the trophoblast was separated from that of the uterine epithelium. It was assumed that the properties of the trophoblast cell membrane make a blastocyst attachment impossible.
