A micromethod is described for the analysis of glycolytic intermediates and high energy phosphates in muscle tissue. The tissue is frozen, sliced to 10—20 thickness in a cryostat, and the slices submitted to ultrasonic disruption. The metabolites are determined fluorimetrically. The composition of the test mixture is given for the following substrates: G-6-P, G-l-P, F-6-P, FDP, DAP, G, M, L, PY, ATP, ADP, AMP, CP and glucose. The values for ATP and CP obtained by this method are only 11 and 19% higher, respectively, than those hitherto obtained by other methods. The values for the glycolytic intermediates are, however, very much lower than those measured previously. The L/P quotient, calculated from the concentrations of L and PY, is about 12; it is thus close to that for other organs and fluids. The low value for glycolytic intermediates and the L/P quotient show that the concentrations determined by the above micromethod approach [ those obtaining in vivo. \ The above method has the following advantages:
