(ƒÀ-heterochromatin was first reported by Heitz (1934), who found in the salivary gland nuclei of Drosophila virilis that the former forms a compact body in contrast to the relatively diffused structure of the latter. This has been made possible by comparing the magnititude and appearance of heterochromatic region shown in the mitotic prophase chromosomes with those of the corresponding parts represented in the salivary gland chromosomes. The works of Muller et al (1937) and of Hinton (1942) on Drosophila melanogaster, and of Pavan (1946) on D. nebulosa have further demonstrated that the j ƒÀ-heterochromatin . contained in a chromo some can be reduced to a single band or to a few chromomeres in the salivary gland chromosome, while a-heterochromatin usually forms the massive chromocenter. Drosophila tumiditarsus (description given in Tan et al, in press) is especially favorable for the study of the distribution of the two kinds of heterochromatin, because unlike other species of Drosophila so far studied, the centromere regions of these long chromosomes are not attached to the chromocenter, making it much easier to distinguish the nature and the amount of heterochromatic material contained in each chromosome. The strain of D. tumiditarsus used in the present study was obtained in Hangchow. The acete-carmine smears of the larval brains and the salivary glands were prepared as usual except the latter were pretreated with HCI (Hsu, 1947).
