Stimulatory feedback action of estradiol (E2) and estrone (E1) on luteinizing hormone (LH) in ovariectomized rats was investigated. A single dose of 20 mcg E2 (primed) caused a 24-hour periodicity of LH secretion that was low at noon and high in the evening. The 2nd dose of E2 given 72 hours later potentiated the LH secretion approximately 2 1/2-folds higher 30 hours after treatment. E1 injection markedly induced an LH increase in rats bearing medial preoptic area (MPO)-roof cut. Both estrogen derivative implants in the suprachiasmatic (POSC) and periventricular portions in the PO area evidently induced an increase of serum LH. The results suggest that the neuronal elements from which the transected fibers in the MPO-roof cut rats originated are the main positive feedback sites for E2 for the induction of the ovulatory LH discharge. Progesterone injection in E2-primed rats as well as progesterone implants in the POSC produced a marked increase in serum LH.
